Ender Backpacks can drop as random loot from killing Bosses (excluding Shrek and Evil Santa). NPCs can theoretically drop an Ender Backpack.

Trivia[]
Ender Backpacks can be enchanted with Curse of Stickiness or Burn Resistance (mcio only). You can do this in an enchanting table; however, you don't always get an enchant option to apply.
